For this question:

Based on the data presented in figures 2 and 3, what is the most likely role of Y229 in protein stability and cAMP activation?


I did not interpret the graphs the right way, so how would I know from the passage that the variants showed they were being removed? I thought it was showing the variant by itself and the effects.

(Since this relates to what is giving you trouble in this question, I do want to emphasize that reading every single character in a figure, graph or passage can be very helpful for the MCAT. As you make a habit of paying attention to details, the notation will start making a bit more sense!)

The problem you are having is not being familiar with the shorthand notation for amino acid substitutions. Y229A means "Replace tyrosine residue at position #229 with alanine". This is very common notation on the MCAT. Both figures talk about this 'Y229A' replacement.
